目的探讨作为转化生长因子β信号通路的核心因子单核细胞趋化蛋白-1(MCP-1)在食管鳞癌(ESCC)中mRNA表达情况,并评价其与ESCC临床及病理特征、预后的关系。方法收集2009年9月至2012年8月外科手术切除的ESCC标本79例,包括鳞癌组织及其配对切缘相对正常食管黏膜组织。采用相对荧光定量RT-PCR检测上述鳞癌和癌旁组织中MCP-1的mRNA水平,据其结果(鳞癌/癌旁)分为3组:高表达组、低表达组和正常表达组。分析3组患者临床及病理特征、患者生存率的差异,从而评估MCP-1mRNA表达与患者临床及病理特征、患者生存率的关系。结果荧光定量RT-PCR结果显示MCP-1基因在食管鳞癌组织中的mRNA水平存在异常表达,高表达43.0%,低表达20.2%;MCP-1的异常表达与瘤块的大小、淋巴结转移相关(P均<0.05),与患者的生存率也密切相关(P<0.05)。结论 MCP-1基因在食管鳞癌组织中的mRNA水平呈异常表达,与食管鳞癌的不良预后密切相关。
Objective To investigate the mRNA expression of monocyte chemoattractant protein-1 (MCP-1), a key factor of transforming growth factor β signaling pathway, in esophageal squamous cell carcinoma (ESCC) and to evaluate its relationship with clinical and pathological features and prognosis of ESCC . Methods Seventy-nine ESCC specimens surgically resected from September 2009 to August 2012 were collected, including squamous cell carcinoma and matched normal esophageal mucosa. The mRNA levels of MCP-1 in squamous cell carcinoma and adjacent tissues were detected by relative fluorescence quantitative RT-PCR. According to the result (squamous cell carcinoma / para-carcinoma), the expression of MCP-1 mRNA was divided into three groups: high expression group, low expression group and normal expression group. The clinical and pathological characteristics of the three groups were analyzed, and the differences in survival rates between the two groups were analyzed to evaluate the relationship between the expression of MCP-1 mRNA and the clinical and pathological features of patients and the survival rate of patients. Results The results of real-time quantitative RT-PCR showed that the mRNA level of MCP-1 was abnormally expressed in esophageal squamous cell carcinoma (43.0%, 20.2%). Aberrant expression of MCP-1 correlated with tumor size and lymph node metastasis (All P <0.05), which were also closely related to the survival rate of patients (P <0.05). Conclusion The mRNA level of MCP-1 gene in esophageal squamous cell carcinoma is abnormally expressed, which is closely related to the poor prognosis of esophageal squamous cell carcinoma.
