我们于1978年11月开始进行按树组织培养育苗试验。1979年1月在冈尼桉的离体茎上诱导出芽;1979年2-6月,先后从葡萄桉和灰桉离体茎诱导发生完整植株,并已进入盆栽试验,生长良好。本文就几种桉树茎段培养中愈伤组织,芽和根的诱导发生情况作简要报道,并进行初步的分析和讨论。材料和方法供试材料取自上海植物园引种栽培的葡萄桉(Eucalyptus botryoides)、灰桉(E.cinerea)、红桉(E.rubida)、冈尼桉(E.gunnii)、银叶山桉(E.pulverulenta)、窿缘桉(E.exserta)、特拉布桉(E.trabuti)、摩洛加桉(E.moluccana)、白木桉(E.leucoxylon)、多枝桉(E.vimiwalis)等。用其2-5年生幼树嫩枝的茎段、茎尖和叶,或从具3-4对真叶的实生苗取其茎段、叶和根作外植体。先在清水中冲洗,后用洗涤粉刷洗,又经清水冲洗后在无菌室内用70%酒精进行表面消毒1分钟,最后用10-15%漂白粉过滤清液消毒20分钟,
We started in November 1978 according to tree tissue culture nursery test. In January 1979, budding was induced on isolated stems of Eucalyptus globulus. From February to June 1979, intact plants were induced in vitro from stems of Eucalyptus urophylla and Eucryptoporullum, and had been in pot experiment and grew well. In this paper, several kinds of eucalyptus stems in callus induction, callus and root induction were briefly reported, and the initial analysis and discussion. MATERIALS AND METHODS The test materials were collected from the Eucalyptus botryoides, E. cinerea, E.rubida, E.gunnii and Eucalyptus globulus cultivated in Shanghai Botanical Garden. E. glutaricum, E. vulgarea, E. exserta, E. trabuti, E. moluccana, E. leucoxylon, E. vimiwalis, etc. . The stem segments, shoot tips and leaves of young shoots of 2- to 5-year-old young shoots, or stems, leaves and roots of 3-4-pairs of true leaves were used as explants.
