目的:研究抗癌药物羟基喜树碱(HCPT)新剂型羟基喜树碱脂质体(LHCPT)在Beagle犬体内的毒物代谢动力学,探讨该药物是否在血液中有蓄积作用。方法:采用HPLC-荧光检测法测定全血中LHCPT的浓度。结果:在第1天和连续给药第30天后Cmax和AUC值存在剂量相关性;药物半衰期t1/2不同剂量之间无显著变化(P>0.05)。AUC、Cmax、t1/2值多次给药与单次给药后比较,同剂量下均未见显著提高(P>0.05)。结论:LHCPT连续静脉给药后未见血液中有明显蓄积现象。
Objective: To study the toxicokinetics of the anti-cancer drug hydroxycamptothecin (HCPT), a new dosage form of hydroxycamptothecin liposome (LHCPT) in Beagle dogs, and explore whether the drug has an accumulation effect in the blood. Methods: The concentration of LHCPT in whole blood was determined by HPLC-fluorescence assay. RESULTS: There was a dose-dependent relationship between Cmax and AUC on the first day and on the 30th day after continuous administration; there was no significant change in the half-life of the drug between t1 / 2 (P> 0.05). AUC, Cmax, t1 / 2 multiple administration and single administration compared with the same dose showed no significant increase (P> 0.05). Conclusion: There is no obvious accumulation of blood in LHCPT after continuous intravenous administration.
