目的:研究中国汉族人群性激素结合球蛋白(Sex hormone-binding globulin,SHBG)基因Asp327Asn多态性与2型糖尿病(Type 2 diabetes mellitus,T2DM)发病风险的关系。方法:应用限制性片段长度多态性(Polymerase chain reaction-restriction fragment length polymorphism,PCR-RFLP)方法,对599例重庆及周边地区汉族人群(T2DM患者248例、正常对照351例)Asp327Asn进行基因分型。用稳态模型(Homeostatic model assessment,HOMA)评估胰岛素抵抗(HOMA-IR)和胰岛β细胞功能(HOMA-B)。结果:Asn等位基因的分布频率T2DM组低于正常对照组(分别为14.5%、20.1%);与Asp等位基因携带者相比,Asn等位基因携带者患T2DM的风险显著降低(OR=0.62,95%CI为0.42~0.91,P=0.016)。结论:SHBG基因Asp327Asn位点Asn等位基因可能与低T2DM患病风险相关。
Objective: To investigate the association between Asp327Asn polymorphism of sex hormone-binding globulin (SHBG) gene and the risk of type 2 diabetes mellitus (T2DM) in Chinese Han population. Methods: The genotypes of Asp327Asn in 599 Han population (T2DM 248 cases, normal control 351 cases) from 599 Chongqing and its surrounding areas were genotyped by PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism) type. Insulin resistance (HOMA-IR) and islet β-cell function (HOMA-B) were assessed using Homeostatic model assessment (HOMA). Results: The frequencies of Asn allele in T2DM group were lower than those in control group (14.5%, 20.1%, respectively). Compared with those with Asp allele, Asn allele had a significantly lower risk of T2DM = 0.62, 95% CI 0.42 to 0.91, P = 0.016). Conclusion: The Asn allele of Asp327Asn site in SHBG gene may be associated with the risk of low T2DM.
