目的 :探讨转录因子激活蛋白2E(transcription factor activator protein 2E,TFAP2E)基因在胃癌组织及正常胃黏膜组织中的甲基化状态及其临床意义。方法 :采用甲基化敏感性高分辨率熔解(methylation-sensitive high-resolution melting,MS-HRM)曲线技术检测90例胃癌组织及31例正常胃黏膜组织中TFAP 2E基因的甲基化水平,分析TFAP2E基因的甲基化水平与胃癌发病和胃癌患者临床病理因素之间的关系。结果 :胃癌组织中TFAP2E基因的甲基化发生率为98.9%(89/90),明显高于正常胃黏膜组织的77.4%(24/31)(P<0.001);TFAP 2E基因的甲基化水平与胃癌的发生呈正相关(r=0.430,P<0.001),受试者工作特征曲线的曲线下面积为0.764,提示TFAP2E基因的甲基化水平对胃癌发病有一定的预测性;TFAP2E基因的甲基化水平与胃癌患者的年龄、性别、病变部位、肿瘤浸润深度、淋巴结转移和远处转移等因素无关(P>0.05),而与胃癌的分化程度和临床分期有关(P<0.001,P=0.006)。结论 :胃癌组织中TFAP2E基因的甲基化发生率高于正常胃黏膜组织,TFAP2E基因的甲基化水平与胃癌的发生呈正相关,胃癌组织中TFAP2E基因的甲基化程度与胃癌的分化程度和临床分期有关。
Objective: To investigate the methylation status of transcription factor activator protein 2E (TFAP2E) gene in gastric carcinoma and normal gastric mucosa and its clinical significance. Methods: Methylation-sensitive high-resolution melting (MS-HRM) curve was used to detect the methylation level of TFAP 2E gene in 90 cases of gastric carcinoma and 31 cases of normal gastric mucosa. Relationship Between Methylation Level of TFAP2E Gene and Clinicopathological Factors in Gastric Cancer and Gastric Cancer. Results: The methylation rate of TFAP2E gene was 98.9% (89/90) in gastric cancer tissues, which was significantly higher than that in normal gastric mucosa (77.4%, 24/31) (P <0.001). The methylation level of TFAP2E is positively correlated with the occurrence of gastric cancer.
