目的:探讨大鼠大脑海马立体定向注射β-淀粉样蛋白1-40(Aβ1-40)对caspase-3蛋白表达的影响及通络救脑口服液的干预作用。方法:140只SD大鼠随机分为空白对照组、假手术组、模型组、盐酸多奈哌齐组、通络救脑口服液低、中、高剂量组(12、24、48mg.kg-1.d-1)共7组。采用大脑海马立体定向注射凝聚态Aβ1-40诱导老年性痴呆动物模型,药物干预4周后,电镜观察海马组织超微结构的变化,实时免疫印迹法检测caspase-3蛋白的表达。结果:电镜观察结果显示:模型组大鼠海马神经元变性,通络救脑口服液能改善大鼠海马神经元病理改变。免疫印迹结果显示:与假手术组比较,模型组大鼠海马组织caspase-3蛋白表达显著升高(P<0.05);与模型组比较,通络救脑口服液组(12、24、48mg.kg-1.d-1)大鼠海马组织caspase-3蛋白表达均显著减少(P<0.05)。结论:海马注射Aβ1-40后大鼠海马组织caspase-3蛋白表达水平明显增高,而通络救脑口服液可以抑制海马组织促凋亡基因caspase-3蛋白的表达,从而发挥其抗老年性痴呆神经细胞凋亡的作用。
Objective: To investigate the effect of β-amyloid 1-40 (Aβ1-40) on the expression of caspase-3 protein in rat hippocampus and the effect of Tongluo Jiuru oral liquid. Methods: One hundred and forty SD rats were randomly divided into control group, sham operation group, model group, donepezil hydrochloride group, Tongluo Jiuru oral liquid low, medium and high dose groups (12,24,48mg.kg-1.d -1) A total of 7 groups. The hippocampus was injected stereotactically with condensed Aβ1-40 to induce the animal model of senile dementia. The changes of ultrastructure in hippocampus were observed by electron microscope and the expression of caspase-3 protein was detected by real-time immunoblotting. Results: Electron microscopy results showed that the degeneration of hippocampal neurons in model group and the improvement of pathological changes of hippocampal neurons in rats by Tongluo Jiunao oral liquid. Western blot results showed that compared with sham operation group, the expression of caspase-3 protein in hippocampal tissue of model group was significantly increased (P <0.05). Compared with model group, Tongluo Jiunao oral liquid group (12,24,48mg. kg-1.d-1) significantly decreased the expression of caspase-3 protein in hippocampus of rats (P <0.05). Conclusion: The expression level of caspase-3 protein in hippocampus of rats hippocampus after Aβ 1-40 injection is significantly increased, while Tongluo Jiunao oral liquid can inhibit the expression of caspase-3 protein in hippocampus, The role of nerve cells in apoptosis.
