目的:建立大剂量化疗时人血浆中甲氨蝶呤浓度的测定方法。方法:以Kromasil C18(4.6mm×250mm,5μm)为色谱柱,流动相为0.05%磷酸盐缓冲液(pH6.8)-甲醇(80-20);柱温45℃;流速0.8mL·min-1;检测波长306nm。结果:在0.08~100mg·L-1范围内,线性关系良好,r=0.9999,最低检测为0.02mg·L-1,平均加样回收率99.2%,日内及日间RSD均小于5%。结论:该方法简单快捷,灵敏度高,结果准确,适用于大剂量化疗时甲氨蝶呤血药浓度监测。
Objective: To establish a method for the determination of methotrexate in human plasma during high-dose chemotherapy. METHODS: Kromasil C18 (4.6 mm × 250 mm, 5 μm) was used as the mobile phase with a mobile phase of 0.05% phosphate buffer (pH 6.8) and methanol (80-20) 1; Detection wavelength 306nm. Results: In the range of 0.08 ~ 100 mg · L-1, the linearity was good, r = 0.9999, the lowest detection was 0.02 mg · L-1, the average recovery was 99.2%. Conclusion: The method is simple, rapid, sensitive and accurate. It is suitable for the monitoring of methotrexate plasma concentration in high-dose chemotherapy.
