目的:建立胆石利通片和胆石利通胶囊中橙皮苷的高效液相色谱测定方法。方法:样品中橙皮苷用甲醇超声提取后,经ZORBAX SB-C18(250 mm×4.6 mm,5μm)分离,色谱柱温度30℃,流动相为甲醇-乙酸-水(35∶4∶61);检测波长283 nm;外标法定量。结果:橙皮苷在24.42 ng~976.8 ng范围内线性关系均良好(r为0.9999),胆石利通胶囊和片剂的方法重复性相对标准偏差分别为1.9%和0.8%;加标回收率分别为99.1%和102.4%;样品溶液在30 h~34 h内稳定性良好。结论:该法简便、灵敏、可靠,可用于胆石利片和胶囊中橙皮苷的质量控制。
Objective: To establish a method for the determination of hesperidin in tanshitong tablet and tanshitong capsule by high performance liquid chromatography. METHODS: The hesperidin was extracted by methanol and separated by ZORBAX SB-C18 (250 mm × 4.6 mm, 5 μm). The chromatographic column temperature was 30 ℃ and the mobile phase consisted of methanol-acetic acid-water (35:4:61) ; Detection wavelength of 283 nm; external standard method. Results: Hesperidin showed a good linear relationship (r = 0.9999) in the range of 24.42 ng to 976.8 ng, and the relative standard deviations (RSDs) of tanshitong capsule and tablet were 1.9% and 0.8%, respectively. 99.1% and 102.4%. The stability of the sample solution was good within 30 h to 34 h. Conclusion: The method is simple, sensitive and reliable and can be used for the quality control of hesperidin in Dan Shili tablets and capsules.
