目的观察鲜鱼腥草挥发油减轻慢性肺损伤及降低趋化因子受体[半胱氨酸-半胱氨酸趋化因子受体2(CCR2)]高表达的作用。方法取SD大鼠,气管滴入脂多糖(LPS)诱发慢性肺损伤模型(模型组))。模型组大鼠给予不同剂量鲜鱼腥草挥发油(给药组;再分为鲜血腥草挥发油高、中、低剂量组)。各组大鼠处死后进行其支气管肺泡灌洗,取其支气管肺泡灌洗液(BALF)离心后沉淀,行WBC计数和WBC分类。取其左肺石蜡切片常规处理,行HE染色、阿尔辛蓝-过碘酸雪夫染色,分析大鼠炎症程度和杯状细胞化生。免疫组织化学染色检测其CCR2的表达。结果 1.模型组大鼠BALF中WBC总数和中性粒细胞数明显高于正常组,肺组织呈不同程度、不同数量的支气管、细支气管慢性炎症。2.经不同剂量鲜鱼腥草挥发油灌胃后能减少WBC总数及中性粒细胞数,降低炎症程度;尤其是鲜鱼腥草挥发油高剂量组。3.给药组大鼠肺泡内CCR2阳性染色区域减少,染色程度减轻;通过图像分析,给药组大鼠肺组织中CCR2的表达较模型组显著降低。结论鲜鱼腥草挥发油可减轻LPS引起的大鼠慢性肺损伤,并能抑制肺泡内CCR2的高表达。
Objective To observe the effect of fresh Houttuynia volatile oil on chronic lung injury and reduce the expression of chemokine receptor [cysteine-cysteine chemokine receptor 2 (CCR2)]. Methods Sprague - Dawley (LPS) - induced chronic lung injury model was induced in SD rats by tracheal instillation. The model group rats were given different doses of fresh Houttuynia flowers volatile oil (administration group; then divided into Houttuynia volatile oil high, medium and low dose group). The rats in each group were sacrificed and their bronchoalveolar lavage was performed. The bronchoalveolar lavage fluid (BALF) was centrifuged and then precipitated. The WBC count and WBC classification were performed. Take the left lung paraffin sections routine treatment, HE staining, Alcian blue - periodic acid Schiff staining, analysis of inflammatory degree and goblet cell metaplasia. Immunohistochemical staining was used to detect the expression of CCR2. The number of WBC and neutrophil in BALF in model group was significantly higher than that in normal group. The lung tissue showed different degrees and different numbers of bronchial and bronchial chronic inflammation. After different doses of fresh Houttuynia volatile oil gavage can reduce the total number of WBC and neutrophils, reduce the degree of inflammation; especially fresh Houttuynia volatile oil high-dose group. The CCR2-positive staining area in the alveolar of the administration group was decreased and the staining degree was alleviated. The CCR2 expression in the lung tissue of the administration group was significantly lower than that of the model group by image analysis. Conclusion Volatile oil from Houttuynia can attenuate chronic lung injury in rats induced by LPS and inhibit the high expression of CCR2 in alveoli.
