目的利用组织芯片检测PI3Kp85α、pAkt、Ki-67蛋白在胃黏膜、胃腺癌及癌旁组织的表达情况,探讨其意义。方法用免疫组化技术检测了30例胃黏膜组织、85胃腺癌组织(分为高、中、低分化3种)及其癌旁组织中PI3Kp85α、pAkt及Ki-67的表达。结果PI3Kp85α、pAkt和Ki-67蛋白在胃黏膜、癌旁组织及胃腺癌组织中的阳性表达率差异有统计学意义(2χ=41.03~57.58,P<0.01),随病情进展三者表达逐步增强(Hc=6.86~13.30,P<0.05、0.01)。在不同分化程度的胃腺癌中PI3Kp85α、pAkt和Ki-67蛋白的阳性表达率差异有显著性(χ2=18.08~18.90,P<0.01),随肿瘤恶性程度增高其阳性表达增强(Hc=15.95~20.10,P<0.01)。PI3Kp85α、pAkt与Ki-67蛋白三者的表达两两相关(r=0.82、0.81、0.86,P<0.01)。结论PI3Kp85α、pAkt和Ki-67的表达与胃腺癌的发生密切相关。
Objective To detect the expression of PI3Kp85α, pAkt and Ki-67 in gastric mucosa, gastric adenocarcinoma and paracancerous tissues by tissue microarray, and to explore its significance. Methods The expressions of PI3Kp85α, pAkt and Ki-67 in 30 gastric mucosa tissues, 85 gastric adenocarcinoma tissues (divided into high, medium and poorly differentiated) and their adjacent tissues were detected by immunohistochemistry. Results The positive expression rates of PI3Kp85α, pAkt and Ki-67 in gastric mucosa, paracancerous tissues and gastric adenocarcinoma tissues were significantly different (2χ = 41.03-57.58, P <0.01), and gradually increased with the progression of the disease (Hc = 6.86 ~ 13.30, P <0.05,0.01). The positive rates of PI3Kp85α, pAkt and Ki-67 in gastric cancer with different degrees of differentiation were significantly different (χ2 = 18.08-18.90, P <0.01). The positive expression of PI3Kp85α, pAkt and Ki- 20.10, P <0.01). The expressions of PI3Kp85α, pAkt and Ki-67 were correlated with each other (r = 0.82,0.81,0.86, P <0.01). Conclusion The expressions of PI3Kp85α, pAkt and Ki-67 are closely related to the occurrence of gastric adenocarcinoma.
