目的观察大剂量激素及木豆叶干预后骨髓间充质干细胞(MSCs)的蛋白质变化,探讨木豆叶治疗股骨头坏死的作用机理。方法培养人MSCs,设正常对照组、模型(激素)组及木豆叶含药血清组,通过双向电泳比较正常对照组与模型组、模型组与木豆叶含药血清组的蛋白质表达差异,通过质谱分析对差异蛋白质进行初步鉴定。结果与正常对照组比较,模型组MSCs有38个蛋白点发生显著变化;与模型组比较,木豆叶含药血清组MSCs有28个蛋白点发生显著变化,MCSc中热休克蛋白(HSP)、含缬络肽蛋白(VCP)及COP9表达水平显著升高。结论大剂量激素干预可造成MSCs内脂肪代谢失调、分化异常,木豆叶可抑制MSCs凋亡,并可能通过MAPK途径,影响其成骨分化及修复骨坏死而发挥治疗股骨头坏死的作用。
Objective To observe the changes of protein in bone marrow mesenchymal stem cells (MSCs) induced by high dose of Hormone and Chaiye leaf intervention and to explore the mechanism of action of Hibiscus cannabinus in treating femoral head necrosis. Methods Human MSCs were cultured, and the normal control group, model group (hormone) group and drug-containing serum group were established. Two-dimensional electrophoresis was used to compare the protein expression differences between the normal control group and the model group, The differential proteins were preliminarily identified by mass spectrometry. Results Compared with the normal control group, there were 38 protein spots in MSCs in the model group significantly changed. Compared with the model group, there were 28 protein spots in the MSCs of the M. officinalis group. The levels of heat shock protein (HSP) VCP and COP9 expression levels were significantly increased. Conclusion High-dose hormones can cause dysregulation of fat metabolism and differentiation in MSCs, which may inhibit the apoptosis of MSCs and may play a role in the treatment of osteonecrosis of the femoral head through MAPK pathway, osteogenesis and osteogenesis.
