目的:探讨广西何首乌中蒽醌类化合物GXHSWAQⅠ,在鼻咽癌CNE-1细胞的放射增敏过程中对乏氧诱导因子表达的影响。方法:MTT比色法确定GXHSWAQⅠ对鼻咽癌CNE-1细胞株体外增敏剂量和放射增敏比,HE染色观察GXHSWAQⅠ放射增敏后细胞形态学的改变,逆转录—聚合酶链反应(RT-PCR)检测GXHSWAQⅠ作用前后,乏氧诱导因子HIF-1α的表达情况。结果:无细胞生长抑制作用浓度的GXHSWAQⅠ低浓度(3.90mg/L)和高浓度(7.80mg/L)组的放射增敏比分别为1.23和1.58,放射和联合用药的细胞发生明显的细胞形态学改变,同时下调基因HIF-1α的表达。结论:GXHSWAQⅠ对鼻咽癌CNE-1细胞具有放射增敏作用,其机制与下调乏氧诱导因子HIF-1α表达有关。
Objective: To investigate the effect of GXHSWAQI, a compound of Radix Polygoni multiflori, on the expression of hypoxia-inducible factors in the radiosensitization of nasopharyngeal carcinoma CNE-1 cells. Methods: MTT colorimetric method was used to determine the sensitizing dose and radiosensitization ratio of GXHSWAQI in nasopharyngeal carcinoma CNE-1 cell line, HE staining was used to observe the change of cell morphology after radiosensitization with GXHSWAQI, and reverse transcription-polymerase chain reaction (RT). The expression of hypoxia-inducible factor HIF-1α was detected before and after GXHSWAQI treatment. RESULTS: The radiosensitization ratios of GXHSWAQI low concentration (3.90 mg/L) and high concentration (7.80 mg/L) group without cell growth inhibitory concentration were 1.23 and 1.58, respectively, and the cell morphology of radiotherapy and combination administration was obvious. Changes in the study, while down-regulating the expression of the gene HIF-1α. CONCLUSION: GXHSWAQI has radiosensitization on CNE-1 cells, and its mechanism is related to down-regulation of HIF-1α.
