目的评价高效液相色谱测定人血清中拉莫三嗪浓度的方法。方法用Hypersil C18色谱柱,柱温为30℃,流动相为1%三(羟甲基)氨基甲烷-乙腈(70∶30),流速为0.8 mL.min-1,测定波长为230 nm。结果拉莫三嗪血清样品线性范围为0.39~50.00 mg.L-1,1.56,6.25,25.00 mg.L-13个浓度质控样品日内、日间精密度(RSD)均<5%,方法学回收率分别为(103.8±3.6)%,(100.0±2.2)%,(99.1±4.2)%。结论本方法回收率高、准确度好、操作简便,适用于临床上对拉莫三嗪血药浓度的常规监测。
AIM To evaluate the method for the determination of lamotrigine in human serum by high performance liquid chromatography. Methods Hypersil C18 column was used. The mobile phase was 1% tris (hydroxymethyl) aminomethane-acetonitrile (70:30) at a flow rate of 0.8 mL · min-1 with a wavelength of 230 nm at 30 ℃. Results The linear range of lamotrigine serum samples was 0.39 ~ 50.00 mg.L-1, 1.56, 6.25 and 25.00 mg.L-13 concentrations of RSD were less than 5% The recoveries were (103.8 ± 3.6)%, (100.0 ± 2.2)% and (99.1 ± 4.2)%, respectively. Conclusion The method of high recovery, good accuracy, easy to operate, suitable for routine monitoring of the blood concentration of lamotrigine.
