目的探讨胃癌组织中葡萄糖调节蛋白78(Grp78)的表达及其与胃癌病理类型、临床分期、分化程度及是否转移的关系。方法胃癌手术切除标本30例,用半定量逆转录-聚合酶链反应(RT-PCR)检测Grp78 mRNA相对表达量;免疫组织化学方法检测Grp78蛋白表达情况。结果半定量RT-PCR显示胃癌组织与正常组织中都有Grp78表达,胃癌组织为1.02±0.16,正常组织为0.62±0.14,差异具有统计学意义。其表达程度与肿瘤大小成正比、随肿瘤分化程度降低而升高,Ⅲ、Ⅳ期患者显著高于I、Ⅱ期,有淋巴结转移者表达程度显著高于无淋巴结转移者,与患者年龄及性别无关。免疫组织化学显示:胃癌组织大多数表达在++~+++,正常组织大多表达在-~++。 Grp78蛋白表达程度与肿瘤大小成正比、随肿瘤分化程度降低而升高,Ⅲ、Ⅳ期患者显著高于I、Ⅱ期,有淋巴结转移者表达程度显著高于无淋巴结转移者。结论胃癌组织的Grp78表达呈增高趋势,参与了胃癌的发展过程,并与胃癌部分生物学行为有关。
Objective To investigate the expression of glucose regulated protein 78 (Grp78) in gastric carcinoma and its relationship with pathological type, clinical stage, differentiation and metastasis of gastric cancer. Methods Thirty patients with gastric cancer underwent surgical resection. The relative expression of Grp78 mRNA was detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). The expression of Grp78 protein was detected by immunohistochemistry. Results Semi-quantitative RT-PCR showed Grp78 expression in both gastric cancer tissues and normal tissues. The gastric cancer tissue was 1.02 ± 0.16 and the normal tissue was 0.62 ± 0.14, the difference was statistically significant. The expression level was proportional to the size of the tumor and increased with the degree of tumor differentiation. The patients in stage Ⅲ and Ⅳ were significantly higher than those in stage I and Ⅱ. The expression of lymph node metastasis was significantly higher than those without lymph node metastasis, Nothing to do Immunohistochemistry showed that the majority of gastric cancer tissues were expressed in ++ ~ +++ and the normal tissues were mostly expressed in ~ ~ ++. Grp78 protein expression is proportional to the size of the tumor, with the degree of tumor differentiation decreased, the patients in stage Ⅲ and Ⅳ were significantly higher than those in stage I and stage Ⅱ, and those with lymph node metastasis were significantly higher than those without lymph node metastasis. Conclusion The expression of Grp78 in gastric cancer is increasing, which is involved in the development of gastric cancer and is related to some biological behaviors of gastric cancer.
