目的探讨Toll样受体TLR 4、7、9 mRNA和蛋白在新疆不同民族食管癌组织中的表达及其与肿瘤浸润和转移之间的关系。方法应用实时荧光定量PCR(qRT-PCR)法检测40例食管癌及相应癌旁组织中TLR4、7、9 mRNA的表达。采用免疫组化SP法检测90例食管癌及相应癌旁组织中TLR4、7、9蛋白的表达。结果 TLR7、9 mRNA在食管癌中的表达量明显高于癌旁组织(P<0.05);TLR4 mRNA在食管癌中的表达量与癌旁组织中的表达,差异无统计学意义(P>0.05)。TLR4、7、9 mRNA在汉族食管癌患者组织中的表达均高于新疆哈萨克族(哈族)(P<0.05)。TLR4、7、9蛋白在食管癌组织中的表达明显高于癌旁组织(P<0.05)。TLR4表达与肿瘤淋巴结转移密切相关(P<0.05);TLR7表达与肿瘤浸润深度相关(P<0.05);TLR4、7、9蛋白表达与肿瘤的分化程度密切相关(P<0.05)。TLR4蛋白在食管癌组织间质炎细胞中高表达与肿瘤的浸润和转移有关,而TLR9蛋白在食管癌组织间质成纤维样细胞中高表达与肿瘤的低浸润和低转移密切相关(P<0.005)。结论TLR4、7、9在食管癌中的高表达与食管癌生物学行为密切相关,提示该基因在食管癌的发生、发展、浸润及转移中起一定作用,其族群差异可能反应不同族群的遗传背景及肿瘤的发病机制。
Objective To investigate the expression of Toll-like receptor TLR 4,7,9 mRNA and protein in esophageal cancer tissues from different nationalities in Xinjiang and its relationship with tumor invasion and metastasis. Methods The expression of TLR4, 7, 9 mRNA was detected in 40 cases of esophageal cancer and its adjacent tissues by real-time quantitative PCR (qRT-PCR). Immunohistochemical SP method was used to detect the expression of TLR4, 7 and 9 protein in 90 cases of esophageal cancer and corresponding adjacent tissues. Results The expression of TLR7 and 9 mRNA in esophageal carcinoma was significantly higher than that in adjacent tissue (P<0.05). There was no significant difference in the expression of TLR4 mRNA between esophageal carcinoma and adjacent tissues (P>0.05). ). The expression of TLR4, 7, 9 mRNA was higher in Han patients with esophageal cancer than in Xinjiang Kazak (Kazakh) (P<0.05). The expression of TLR4, 7 and 9 protein in esophageal cancer tissue was significantly higher than that in adjacent tissue (P<0.05). TLR4 expression was closely related to tumor lymph node metastasis (P<0.05); TLR7 expression was correlated with tumor invasion depth (P<0.05); TLR4, 7, 9 protein expression was closely related to tumor differentiation (P<0.05). The high expression of TLR4 protein in esophageal carcinoma tissues was related to tumor invasion and metastasis. The high expression of TLR9 protein in fibroblast-like cells of esophageal carcinoma was closely related to low invasion and low metastasis of tumor (P<0.005). . Conclusion The high expression of TLR4, 7 and 9 in esophageal cancer is closely related to the biological behavior of esophageal cancer, suggesting that this gene may play a role in the occurrence, development, invasion and metastasis of esophageal cancer.
