目的:建立大鼠长春瑞滨外渗性皮肤损伤的理想实验模型,在此动物模型上研究复脉膏外敷对外渗性损伤皮肤中EGFR的影响,以探讨复脉膏外敷治疗化疗性静脉炎的疗效及机制。方法:42只大鼠随机分为两组:复脉膏组、50%硫酸镁组,21只/组,建立实验模型。两组大鼠左侧损伤处分别予复脉膏外敷、50%硫酸镁湿敷;右侧予生理盐水外敷作为对照组。在损伤后第1、4、7、11、14、18天,分别随机取3只大鼠,切除损伤部位的皮肤及皮下组织,固定标本,脱水,石蜡包埋,连续切片,采用免疫组化标记及图像分析方法,在电子显微镜下观察EGFR的表达情况。结果:EGFR阳性表达主要见于表皮细胞、真皮成纤维细胞、毛囊上皮细胞、血管内皮细胞、巨噬细胞、中性粒细胞的胞膜及胞浆中。复脉膏外敷较硫酸镁湿敷及生理盐水外涂能更早更强刺激长春瑞滨外渗性损伤皮肤中EGFR的阳性表达(P<0.05)。结论:复脉膏外敷于损伤皮肤后能显著促进EGFR的快速增加来促进肉芽组织及鳞状上皮的生成,进而促进损伤皮肤的愈合。因此,复脉膏可以用于防治化疗性静脉炎。
OBJECTIVE: To establish an ideal experimental model of vinblastine-induced skin lesions in rats. To study the effect of Fumai cream on EGFR in skin of extravasation injury in animal model to explore the effect of Fumai Paste on the treatment of chemotherapy phlebitis Efficacy and mechanism. Methods: Forty-two rats were randomly divided into two groups: Fumai cream group, 50% magnesium sulfate group and 21 / group. On the left side of the rats in the two groups, they were given plaster externally and 50% magnesium sulphate respectively. The right side was treated with saline as the control group. On the 1st, 4th, 7th, 11th, 14th and 18th days after injury, 3 rats were randomly selected, the skin and subcutaneous tissue were excised at the site of injury, the specimens were fixed, dehydrated and embedded in paraffin, Marker and image analysis methods, under the electron microscope to observe the expression of EGFR. Results: The positive expression of EGFR mainly found in the epidermal cells, dermal fibroblasts, hair follicle epithelial cells, vascular endothelial cells, macrophages, neutrophils in the cytoplasm and cytoplasm. Fumai plaster external application compared with magnesium sulfate wet coating and saline applied earlier and stronger stimulation of vinorelbine skin injury EGFR expression (P <0.05). Conclusion: Fumai cream topical application on the skin can significantly promote the rapid increase of EGFR to promote the formation of granulation tissue and squamous epithelium, and then promote the healing of damaged skin. Therefore, Fumai cream can be used to control chemotherapy phlebitis.
