以葡萄生单轴霉病原菌的人工保存和培养为目的,采用寄主植物和病原菌离体双培养的方法,确定了葡萄生单轴霉离体双培养用的葡萄品种、影响葡萄苗生长的最佳生长调节剂浓度,并建立了离体双培养的操作体系。试验结果表明葡萄无菌苗的最适继代培养基为MS+BA 0.5 mg/L+IBA 0.15 mg/L;不定芽分化培养基为1/2MS+BA 0.3mg/L+IBA 0.1mg/L;最适生根培养基为1/2MS+NAA 0.1mg/L。
