目的 探讨咖啡酸苯乙酯(caffeicacidphenethylester ,CAPE)对体外培养的大肠癌细胞HCT116细胞周期和cyclinD1表达的影响。方法 以人大肠癌细胞株HCT116为研究对象,PI染色、流式细胞仪检测CAPE处理2 4h后细胞周期分布,Westernblotting、RT PCR检测CAPE处理2 4、48h后cyclinD1表达的变化。结果 2 5、5 0、10mg LCAPE处理HCT116细胞2 4h后,G0 G1 期细胞百分率上升,S期细胞百分率下降,呈剂量依赖性。2 5、5 0、10mg LCAPE处理HCT116细胞2 4、48h后cyclinD1蛋白和mRNA表达均降低,呈剂量和时间依赖性。结论 CAPE诱导HCT116细胞周期阻滞,其机制可能与其下调cyclinD1表达有关。
Objective To investigate the effect of caffeic acid polyphenylene ether (CAPE) on the cell cycle and cyclinD1 expression in cultured human colorectal cancer cell line HCT116. Methods The human colorectal cancer cell line HCT116 was used as the research object. The cell cycle distribution was detected by PI staining and flow cytometry after 24 h CAPE treatment. The cyclinD1 expression was detected by Western blotting and RT PCR at 2, 48 hours after CAPE treatment. Results After treated with 2, 5, 10 and 10 mg LCAPE for 24 h, the percentage of cells in G0 G1 phase increased and the percentage of S phase cells decreased in a dose-dependent manner. The expression of cyclinD1 protein and mRNA in HCT116 cells treated with 2, 5, 10 and 10 mg LCAPE for 2 and 48 hours decreased in a dose and time dependent manner. Conclusions CAPE induces cell cycle arrest in HCT116 cells, which may be related to its down-regulation of cyclinD1 expression.
