目的探究环境类雌激素邻苯二甲酸二丁酯(DBP)诱导建立小鼠隐睾模型的方法及其作用机制。方法将7~8周龄的BALB/c怀孕小鼠40只随机分为对照组(n=10),DBP低剂量诱导组(n=10),DBP中剂量诱导组(n=10),DBP高剂量诱导组(n=10),在怀孕的第12天开始实验组填喂DBP,剂量分别为100mg·kg-1·d-1、300mg·kg-1·d-1、500mg·kg-1·d-1,对照组填喂100mg·kg-1·d-1的玉米油,计算新生小鼠睾丸到膀胱颈的距离与肾下极到膀胱颈距离的比值,并取睾丸组织检测NO合酶(NOS)活性和Caspase-3蛋白水平变化。结果经DBP诱导后,小鼠睾丸到膀胱颈的距离与睾丸到肾下极距离的比值随着DBP浓度的增加显著增大(P<0.05);对照组、低剂量组、中剂量组和高剂量组NOS活性分别为(0.37±0.12)nmol·min-1·g-1、(0.59±0.09)nmol·min-1·g-1、(0.98±0.11)nmol·min-1·g-1、(1.35±0.07)nmol·min-1·g-1,且随着剂量的增加NOS活性显著增加(P<0.05);低剂量组、中剂量组和高剂量组Caspase-3蛋白活化程度分别为1.31±0.36、01.89±0.68、2.71±0.39,且随着剂量的增加Caspase-3蛋白活化程度显著加强(P<0.05)。结论环境类雌激素DBP能够明显诱导小鼠隐睾模型的建立,并显著改变睾丸组织的NOS活性和Caspase-3活化程度。
Objective To investigate the method and mechanism of environmental estrogen dibutyl phthalate (DBP) induced mouse cryptorchid model. Methods Forty BALB / c pregnant mice aged 7-8 weeks were randomly divided into control group (n = 10), DBP low dose group (n = 10), DBP medium dose group (n = 10), DBP In the high-dose induction group (n = 10), DBP was fed to the experimental group on the 12th day of pregnancy with doses of 100 mg · kg-1 · d-1, 300 mg · kg-1 · d-1 and 500 mg · kg- 1 · d-1. Conclusion Environmental estrogen DBP can significantly induce the establishment of cryptorchidism in mice and significantly change the activity of NOS and the activation of Caspase-3 in testis.
