目的:探讨红花黄色素A对盲肠结扎穿孔(cecalligation-perforation,CLP)模型诱导的大鼠早期凝血功能及炎症反应的作用。方法:健康SD大鼠120只分4组:对照组、假手术组、CLP组(模型组)、CLP+红花黄色素A组,观察红花黄色素A对CLP模型诱导的大鼠脓毒症时凝血功能的影响及血清中组织因子(tissue factor,TF)及组织因子途径抑制物(tissue factorpathway inhibitor,TFPI)、肿瘤坏死因子-α(tumor necrosis factor-a,TNF-α)、白介素-1β(interleukin-1β)的变化。结果:CLP组与对照组、假手术组相比较,凝血酶原时间(prothrombin time,PT)、活化部分凝血激酶时间(activated partial thromboplastin time,APTT)水平均增加(P<0.05),CLP+红花黄色素A组PT及APTT水平较CLP组降低(P<0.05)。CLP组IL1-β、TNF-α、TF、TFPI水平相较于对照组、假手术组均升高(P<0.05),CLP+红花黄色素A组IL1-β、TNF-α、TF水平较CLP组增加,TFPI水平较CLP组降低(P<0.05)。结论:红花黄色素A除了可以改善凝血异常也可以降低炎性反应,安全、有效、易获取,可成为脓毒症早期治疗及缓解脓毒症时的临床辅助药物。
Objective: To investigate the effect of safflower yellow A on early coagulation and inflammation induced by cecalligation-perforation (CLP) model in rats. Methods: One hundred and twenty healthy SD rats were divided into 4 groups: control group, sham operation group, CLP group (model group) and CLP + safflower yellow group A. The effects of safflower yellow A on CLP model induced sepsis in rats (TFPI), tumor necrosis factor-α (TNF-α), interleukin-1β (interleukin-1β) changes. Results: The levels of prothrombin time (PT) and activated partial thromboplastin time (APTT) in CLP group were significantly higher than those in control group and sham operation group (P <0.05) The levels of PT and APTT in yellow dye A group were lower than those in CLP group (P <0.05). The levels of IL1-β, TNF-α, TF and TFPI in CLP group were significantly higher than those in control group and sham-operated group (P <0.05) CLP group increased, TFPI levels were lower than the CLP group (P <0.05). Conclusion: Safflower A can not only improve the abnormal blood coagulation, but also reduce inflammatory reaction, which is safe, effective and easy to obtain. It can be used as clinical adjuvant drugs in the early treatment of sepsis and sepsis.
