建立了一种高效液相色谱法快速测定血清维生素C含量的方法。Shim-pack VP-ODS C18色谱柱,流动相为0.05 mol/L KH2PO4,pH值为3.0,流速为1 mL/min,检测波长为254 nm。血清样品经50 g/L偏磷酸溶液提取。结果表明,维生素C在浓度为0.5~25μg/mL范围内有良好的线性关系,线性回归方程为Y=(2.429 227×10-5)X+(-8.645 801×10-2),相关系数为0.999 1,方法回收率为98%~102%,相对标准偏差小于3%。本测定方法简便,快速,准确,稳定。
A rapid method for the determination of serum vitamin C content was established by high performance liquid chromatography. Shim-pack VP-ODS C18 column with a mobile phase of 0.05 mol / L KH2PO4 at a pH of 3.0 at a flow rate of 1 mL / min and a detection wavelength of 254 nm. Serum samples were extracted by 50 g / L meta-phosphoric acid solution. The results showed that there was a good linear relationship between the concentration of vitamin C and the concentration of 0.5-25 μg / mL. The linear regression equation was Y = (2.429 227 × 10-5) X + (- 8.645 801 × 10-2) with a correlation coefficient of 0.999 1, the recovery rate was 98% ~ 102%, the relative standard deviation was less than 3%. The determination method is simple, fast, accurate and stable.
