目的 建立反相高效液相色谱法测定四逆散抗抑郁有效部位中新橙皮苷含量的方法。方法选用YWG C18色谱柱 (2 5 0mm× 4 6mm ,10 μm) ,乙腈 - 0 1%磷酸水溶液 (2 0∶80 )为流动相 ,检测波长为 2 83nm ,柱温 :室温 ,对新橙皮苷进行含量测定。结果 新橙皮苷进样量在 0 0 816~ 0 4 896 μg范围内与其峰面积积分值呈良好的线性关系 ,r =0 9995 ,平均加样回收率为 97 10 % ,RSD =0 86 %(n =5 )。结论 本方法简便、准确、可靠 ,可作为四逆散抗抑郁有效部位质量控制的方法。
Objective To establish a reversed-phase high performance liquid chromatographic method for determination of neohesperidin in anti-depressant effective fraction of Sini San. Methods YWG C18 column (250 mm × 46 mm, 10 μm) was used. Acetonitrile- 0 1% phosphoric acid aqueous solution (20:80) was used as the mobile phase. The detection wavelength was 283 nm. Column temperature: room temperature. The glycosides were assayed for content. Results There was a good linear relationship between the injection quantity of neohesperidin and its peak area in the range of 0 0 816 to 0 4 896 μg, r =0 9995, the average recovery was 97 10 %, RSD = 0 86 %. (n =5). Conclusion This method is simple, accurate and reliable. It can be used as a method to control the quality of effective anti-depressant sites in Sini San.
