目的研究贯郁胶囊对嗅球损毁大鼠行为及下丘脑-垂体-肾上腺(HPA)轴的影响。方法采用敞箱法、跳台法观察嗅球损毁大鼠的行为;采用放免法分析贯郁胶囊对嗅球损毁模型大鼠血浆促肾上腺皮质激素(ACTH)和皮质醇(CORT)含量的影响。结果嗅球损毁大鼠敞箱实验的爬格数和站立数显著增加,跳台实验的错误次数明显增多(P<0.01);血浆ACTH和CORT含量明显增高(P<0.01)。贯郁胶囊1.2、0.6g/kg均可降低嗅球损毁大鼠敞箱实验的爬格数和站立数,减少跳台实验的错误次数(P<0.01,P<0.05);贯郁胶囊1.2、0.6、0.3g/kg均能使血浆ACTH和CORT含量明显低于模型组(P<0.05)。结论贯郁胶囊能纠正嗅球损毁大鼠的行为改变,而且对亢进的HPA轴功能的调整作用是贯郁胶囊治疗抑郁症的机制之一。
Objective To study the effect of Guanyu Capsule on the behavior of rats and the hypothalamic-pituitary-adrenal (HPA) axis in the olfactory bulb lesions. Methods The behaviors of rats damaged by olfactory bulb were observed using the open box method and the method of step-down. The effects of guanyu capsule on plasma ACTH and cortisol (CORT) levels in rats with olfactory bulb lesion were analyzed by radioimmunoassay. Results The olfactory bulb damage rat open box experiment significantly increased the number of crawling and standing, the number of errors in the jumping platform experiment increased significantly (P<0.01), and the plasma ACTH and CORT content increased significantly (P<0.01). Both Guanyu capsules (1.2 and 0.6 g/kg) can reduce the number of crawls and standings in rats exposed to the olfactory bulb in the open box experiment, and reduce the number of errors in the stepping experiments (P<0.01, P<0.05); Guanyu capsules 1.2, 0.6, 0.3g/kg can make plasma ACTH and CORT levels significantly lower than the model group (P <0.05). Conclusion Guanyu capsule can correct the behavior change of rats with olfactory bulb damage, and the adjustment of the function of hyperactive HPA axis is one of the mechanisms of Guanyu Capsule in the treatment of depression.
