照射后淋巴样细胞发生迅速的间期死亡,这一过程至少含有两个主要方面,即核质的破坏和胞膜的损伤。本文在γ线体外照射后6小时内,比较几个依赖于照射剂量胸腺细胞损伤的细胞学指标。实验取同窝雌性大鼠,断头处死,收集全血样本于含肝素的玻璃管中,用生理盐水反复离心,去除白细胞层,提纯红细胞(RBC)。剪碎胸腺,网沙过滤,用含10%新鲜大鼠血清的199培养液稀释为5×10~6细胞/毫升,进行钴源照射,剂量率为1.12戈瑞/分,照射后37℃水浴孵育。测定方法如下:
Rapid interphase death of lymphoid cells occurs after irradiation. This process has at least two major aspects, namely nuclear destruction and membrane damage. In this study, several cytological indicators that depend on irradiation doses of thymocyte injury were compared within 6 hours after gamma irradiation. Female littermates were used in the experiment and were sacrificed by decapitation. Whole blood samples were collected in heparin-containing glass tubes and repeatedly centrifuged with physiological saline to remove leukocyte layers and purify red blood cells (RBCs). The thymus was dissected and the mesh was filtered. The cells were diluted with 199 medium containing 10% fresh rat serum to 5×10~6 cells/ml. The cobalt source was irradiated at a dose rate of 1.12 Gy/min and the water bath was 37°C after irradiation. Incubate. The method of measurement is as follows:
