本项研究用五种野生灵芝提取物混合液(口服液),用9g/kg/d的剂量,给接种人鼻咽低分化鳞状上皮癌细胞株(每只棵鼠接种2×10 ̄6细胞数)3天后的裸鼠灌胃给药,对照组用生理盐水20ml/kg/d,两组均按上述剂量分别灌胃20天,裸鼠饲养于温度25℃。相对温度为50%~70%的无菌室内。定期观察各组裸鼠的生长及成瘤情况,实验结果时将动物处死,称瘤重、鼠重;并计算其抑瘤率,取瘤组织肺、肝做组织切片。结果表明五色灵芝口服液对人鼻咽癌细胞株在裸鼠成瘤有明显的抑制作用,经三次试验证实肿瘤的抑制率在38.5~44.2%之间,均P<0.001,各组裸鼠用药后体重均有较明显增加,其增长幅度与对照组相比基本无差异,各组小鼠在实验中一般状态良好,在本实验的条件下未见本药有毒副作用。
In this study, five wild ganoderma lucidum extracts (oral liquid) were used to inoculate human nasopharyngeal poorly differentiated squamous cell carcinoma cell lines at a dose of 9 g/kg/day (2 × 10 6 cells per mouse). Three days later, nude mice were intragastrically administered, and saline (20 ml/kg/d) was administered to the control group. Both groups were intragastrically administered at the above dose for 20 days. Nude mice were maintained at a temperature of 25°C. The relative temperature is 50% to 70% in a sterile room. The growth and tumorigenicity of each group of nude mice were observed regularly. When the experimental results were taken, the animals were sacrificed and the tumor weight and mouse weight were weighed; the tumor inhibition rate was calculated and the tumor tissue lungs and liver were taken for tissue sectioning. The results showed that Wuxing Lingzhi Oral Liquid had a significant inhibitory effect on human nasopharyngeal carcinoma cell lines in nude mice. The inhibition rate of tumors was 38.5-44.2% after three tests, all P<0.001. The body weight of each group of nude mice significantly increased after drug administration. The growth rate was basically the same as that of the control group. The mice in each group were generally in good condition during the experiment. Under the conditions of this experiment, no toxic side effects were observed.
