目的:建立一种海参体壁中总皂苷的含量测定方法,比较我国11种不同种类市售海参和8批格皮氏海参干燥体壁中的总皂苷含量。方法:采用紫外-可见分光光度法,以齐墩果酸为对照品建立标准曲线,5%香草醛-冰醋酸为显色剂,检测波长546 nm,比色法测定总皂苷含量。结果:所建立的方法稳定可靠,不同种海参体壁皂苷含量差异较大,格皮氏海参总皂苷含量最高,市售格皮氏海参皂苷含量在3.16%~5.27%之间。结论:方法简便、准确、重复性好,可用于对海参皂苷的含量测定。对我国大部分市售海参种类皂苷含量进行了测定,其中格皮氏海参可以作为一种很好的制备海参总皂苷的原料药材。
OBJECTIVE: To establish a method for the determination of total saponins in the body wall of sea cucumber, and to compare the content of total saponins in 11 kinds of commercially available sea cucumbers and 8 batches of dry skin of sea cucumber in China. Methods: UV-Vis spectrophotometry was used to establish a standard curve with oleanolic acid as the reference substance. The standard curve was established with 5% vanillin-glacial acetic acid and the detection wavelength was 546 nm. The content of total saponins was determined by colorimetry. Results: The established method was stable and reliable. The content of bulk saponin in different species of sea cucumber was quite different. The content of total saponin in sea cucumber was the highest. The content of Ginsenosides in commercial sea cucumber was between 3.16% and 5.27%. Conclusion: The method is simple, accurate, reproducible and can be used to determine the content of ginsenosides. Most of the commercially available sea cucumber species saponin content was measured, which can be used as a good Geopark sea cucumber ginseng saponins of raw materials.
