目的:多指标综合评分法优选参连方的水提工艺,为参连颗粒的生产提供工艺参数。方法:将大鼠随机分为空白组,提取物Ⅰ组(水提),提取物Ⅱ组(水提醇沉),提取物Ⅲ组(党参等水提醇沉,丹参等醇提),提取物Ⅳ组(党参等水提,丹参等醇提),在以大鼠心肌缺血再灌注后室性早搏、室性心动过速和室性颤动的总时间为指标考察不同提取工艺路线的基础上,采用HPLC测定芍药苷、甘草苷和甘草酸的含量,流动相乙腈(A)-0.1%磷酸水溶液(B)梯度洗脱(0~18 min,86%B;18~30 min,86%~72%B;30~40 min,72%~65%B;40~50 min,65%~60%B;50~55 min,60%~40%B;55~60 min,40%~86%B;60~65 min,86%B),流速1.0 mL·min~(-1),芍药苷检测波长230 nm,甘草苷和甘草酸检测波长237 nm。通过正交试验考察溶剂用量、提取时间、提取次数对3种成分含量的影响,利用综合评分法优选提取工艺并进行验证试验。结果:最佳提取工艺为提取3次,分别加10,8,8倍量水提取1.5 h;芍药苷、甘草苷、甘草酸提取量分别为22.88,15.17,13.54 mg·g~(-1)。结论:优选的提取工艺稳定可行,可为参连颗粒的生产提供指导。
OBJECTIVE: The multi-index comprehensive grading method is optimized for the water extraction process of ginseng, providing process parameters for the ginseng production. Methods: The rats were randomly divided into blank group, extract Ⅰ group (water extract), extract Ⅱ group (water extract alcohol sink), extract group Ⅲ (Codonopsis such as water alcohol precipitation, Salvia ethanol extraction), extraction Group IV (Codonopsis such as water extraction, salvia ethanol extraction), in rat myocardial ischemia-reperfusion after premature ventricular contractions, ventricular tachycardia and ventricular fibrillation of the total time as an index to examine the different extraction process based on the The contents of paeoniflorin, glycyrrhizin and glycyrrhizic acid were determined by HPLC. The mobile phase consisted of gradient elution with acetonitrile (A) -0.1% phosphoric acid (B) (0-18 min, 86% B; 72% -65% B; 40-50 min, 65-60% B; 50-55 min, 60-40% B; 55-60 min, 40-86% B; 60-65 min, 86% B) at a flow rate of 1.0 mL · min -1. The detection wavelength of paeoniflorin was 230 nm. The detection wavelength of glycyrrhizin and glycyrrhizin was 237 nm. The effects of solvent dosage, extraction time and extraction times on the content of three components were investigated by orthogonal test. The extraction process was optimized by comprehensive score method and the validation test was carried out. Results: The optimal extraction process was extraction for 3 times, respectively, for 10 hours. The extraction of paeoniflorin, glycyrrhizin and glycyrrhizic acid were respectively 22.88,15.17 and 13.54 mg · g -1 . Conclusion: The optimal extraction process is stable and feasible, which can provide guidance for the production of ginseng granules.
