目的 观察血管形成抑制剂TNP - 4 70与化疗药物阿霉素联合应用抑制小鼠膀胱癌的协同作用。方法 分别予TNP - 4 70、阿霉素、TNP - 4 70 +阿霉素治疗T739膀胱癌荷瘤小鼠 ,观察肿瘤生长及血管形成、细胞凋亡情况。结果 治疗后 12d ,TNP - 4 70、阿霉素及TNP - 4 70 +阿霉素治疗组抑瘤率分别为 4 6 .3%、2 1.4 %及 5 6 .5 % ,TNP - 4 70组微血管密度明显减少、凋亡指数增多 ,阿霉素组凋亡指数增多 ,TNP - 4 70+阿霉素组凋亡指数进一步增加。结论 TNP - 4 70与阿霉素治疗小鼠膀胱肿瘤 ,有明显的抗肿瘤协同作用 ,其机理可能是通过抑制血管形成及化疗药物细胞毒性作用而促使肿瘤细胞凋亡进一步增加。
Objective To observe the synergistic effect of combination of TNP - 4 70, an inhibitor of angiogenesis, with doxorubicin in inhibiting mouse bladder cancer. Methods T739 bladder cancer bearing mice were treated with TNP - 4 70, doxorubicin and TNP - 4 70 + doxorubicin respectively. The growth, angiogenesis and apoptosis were observed. Results On the 12th day after treatment, the inhibitory rates of TNP - 4 70, doxorubicin and TNP - 4 70 + doxorubicin were 46.3%, 21.4% and 56.5%, respectively, and TNP - 4 70 Microvessel density was significantly decreased, apoptosis index increased, apoptosis index increased in doxorubicin group, apoptosis index in TNP - 4 70+ adriamycin group increased further. Conclusions TNP - 4 70 and doxorubicin have obvious anti - tumor synergistic effects on bladder cancer in mice. The mechanism may be that the apoptosis of tumor cells may be further increased by inhibiting the formation of blood vessels and cytotoxicity of chemotherapy drugs.
