目的探讨肿瘤易感基因TSG101(Tumor susceptibility gene 101)及p21蛋白在胃腺癌中的表达及其临床意义。方法应用免疫组织化学方法检测80例胃腺癌及对应的癌旁2 cm胃黏膜组织中TSG101和p21蛋白的表达情况,结合临床资料分析其临床病理意义。结果①在胃腺癌组织及对应的癌旁2 cm胃黏膜组织中,TSG101蛋白的阳性表达率分别为35.0%和65.0%,p21蛋白阳性表达率分别为32.5%和67.5%,其差异有统计学意义(P<0.05);②TSG101蛋白阳性表达率在胃腺癌的高中分化组及无淋巴结转移组的阳性率为46.2%和67.6%,高于低分化组(14.3%)及有淋巴结转移组(10.9%),其差异均有统计学意义(P<0.01);而在不同TNM分期之间,则差异无统计学意义(P>0.05);③p21蛋白阳性表达率在高中分化组为44.2%,高于低分化组的10.7%,在无淋巴结转移组及TNMⅠ~Ⅱ组,p21蛋白阳性率分别为61.8%和41.8%,高于有淋巴结转移组及TNMⅢ组,组间差异均有统计学意义(P<0.01);④TSG101与P21蛋白之间未见相关性。结论 TSG101可能作为一个新的抑癌基因参与胃癌的发生发展过程,检测TSG101和P21蛋白的表达有助于判定胃腺癌的生物学行为。
Objective To investigate the expression of tumor susceptibility gene (TSG101) and p21 protein in gastric adenocarcinoma and its clinical significance. Methods The expressions of TSG101 and p21 protein in 80 cases of gastric adenocarcinoma and their adjacent 2 cm gastric mucosa were detected by immunohistochemistry. The clinicopathological significance was analyzed based on clinical data. Results ① The positive expression rates of TSG101 protein in gastric adenocarcinoma tissues and their adjacent 2 cm gastric mucosa tissues were 35.0% and 65.0%, respectively, and the positive rates of p21 protein expression were 32.5% and 67.5%, respectively (P <0.05). (2) The positive rates of TSG101 protein expression in high and middle differentiation group and non-lymph node metastasis group were 46.2% and 67.6%, respectively, which were higher than those in poorly differentiated group (14.3%) and lymph node metastasis group %), The differences were statistically significant (P <0.01), but in different TNM stages, the difference was not statistically significant (P> 0.05); ③p21 protein positive expression rate in high school group was 44.2%, high In 10.7% of poorly differentiated group, the positive rates of p21 protein in non-lymph node metastasis group and TNMⅠ ~ Ⅱ group were 61.8% and 41.8%, respectively, which were higher than those in lymph node metastasis group and TNMⅢ group P <0.01); ④ There was no correlation between TSG101 and P21 protein. Conclusion TSG101 may be involved in the carcinogenesis and progression of gastric cancer as a new tumor suppressor gene. Detection of the expression of TSG101 and P21 proteins may be helpful to judge the biological behavior of gastric adenocarcinoma.
