目的:研究昆仙胶囊对胶原诱导关节炎大鼠滑膜组织白细胞介素-8(IL-8)及干扰素-γ诱导蛋白10(γIP-10)mRNA表达的影响。方法:将54只SPF级Wistar大鼠随机分为正常组、模型组、甲氨喋呤(MTX)组和昆仙胶囊低、中、高剂量组各9只。采用尾根部皮下注射胶原蛋白建立胶原诱导关节炎大鼠模型,检测各组关节滑膜细胞与软骨的病理变化,R T-PCR法检测各组滑膜组织IL-8及γIP-10 mRNA的表达水平。结果:MTX组和昆仙胶囊各剂量组大鼠关节滑膜损伤较模型组明显改善(P<0.05,P<0.01)。与模型组比较,MTX组、昆仙胶囊各剂量组大鼠的IL-8 mRNA、γIP-10 mRNA的表达明显降低(P<0.05,P<0.01);随着昆仙胶囊剂量的增加,关节滑膜组织IL-8 mRNA、γIP-10 mRNA的表达有下降的趋势;与MTX组、昆仙胶囊低、中剂量组大鼠比较,昆仙胶囊高剂量组大鼠的IL-8 mRNA、γIP-10 mRNA的表达明显降低(P<0.05)。结论:昆仙胶囊可能通过抑制大鼠滑膜组织IL-8及γIP-10 mRNA基因表达,减轻大鼠滑膜细胞增生及软骨破坏来实现其治疗滑膜炎的目的。
Objective: To investigate the effect of Kunxian capsule on the expression of interleukin-8 (IL-8) and interferon-γ-inducible protein 10 (γIP-10) mRNA in synovial tissue of collagen-induced arthritis rats. Methods: Fifty-four SPF Wistar rats were randomly divided into normal group, model group, methotrexate (MTX) group and Kunxian capsule low, medium and high dose groups of 9 rats. Collagen-induced arthritis rat model was established by subcutaneous injection of collagen into the root of the tail. The pathological changes of synovial cells and cartilage in each group were detected. The expression of IL-8 and γIP-10 mRNA in synovial tissue were detected by RT-PCR Level. Results: Compared with the model group, the synovial injury of MTX group and Kunxian capsule dose groups were significantly improved (P <0.05, P <0.01). Compared with the model group, the expressions of IL-8 mRNA and γIP-10 mRNA in the MTX group and Kunxian capsule groups were significantly decreased (P <0.05, P <0.01). With the increase of the dose of Kunxian capsule, The expression of IL-8 mRNA andγIP-10 mRNA in synovial tissue tended to decrease. Compared with MTX group and Kunxian capsule low and middle dose groups, the expression of IL-8 mRNA, γIP -10 mRNA expression was significantly lower (P <0.05). Conclusion: Kunxian capsule may achieve its goal of treating synovitis by inhibiting the expression of IL-8 and γIP-10 mRNA in synovial tissue of rats and alleviating the synovial cell proliferation and cartilage destruction in rats.
