目的:获得金银花四倍体植株。方法:离体条件下应用秋水仙素浸泡法和混合培养法对金银花愈伤组织进行了不同浓度和时间的处理。结果:以0.1%秋水仙素浸泡15 h诱导效果最佳,诱变率为30.5%。茎尖染色体压片表明,四倍体染色体数为2n=4x=36,二倍体对照为2n=2x=18。获得的金银花四倍体与二倍体植株相比,四倍体植株形态特征变化明显,植株变粗壮,叶片变厚,颜色变深,茎基部粗壮,生长势变缓。气孔、保卫细胞明显增大,气孔密度明显减小。结论:秋水仙素浸泡法可以作为金银花多倍体的诱导方法。
Objective: To obtain tetraploid plants of honeysuckle. Methods: In vitro, colchicine infusion method and mixed culture method were used to treat callus in different concentration and time. Results: Soaking with 0.1% colchicine for 15 h resulted in the best induction rate with a mutation rate of 30.5%. The tip chromosome chromosomes showed that the number of tetraploid chromosomes was 2n = 4x = 36, and the diploid control was 2n = 2x = 18. Compared with diploid plants, tetraploid tetraploid plants obtained obvious changes in their morphological characteristics. The plants became thick and thick, the leaves became thickened, the color became dark, and the stem base was stout and the growth potential became slow. Stomata, guard cells increased significantly stomatal density decreased significantly. Conclusion: Colchicine soaking method can be used as polyploid induction method.
