目的观察牛磺酸联合葛根素对2型糖尿病大鼠肾脏的保护作用。方法将成模的40只Wistar大鼠随机分为糖尿病组(DM组)、单用牛磺酸组(Tau组)、单用葛根素组(Pue组)和牛磺酸联合葛根素组(Tau+Pue组)。8周后,测4组大鼠血浆葡萄糖(Glucose)、肌酐(Cre)、尿素氮(BUN)、超氧化物歧化酶(SOD)、丙二醛(MDA)、活性氧(ROS)、胰岛素(Insulin)及肾脏组织MDA、ROS及SOD,计算大鼠肾指数,光镜下观察肾脏的病理形态学变化。结果与DM组大鼠比较,Tau组、Pue组及Tau+Pue组血浆SOD活性和胰岛素水平升高,而Glucose、Cre、BUN、MDA、ROS和肾指数呈不同程度的降低,并且3个给药组大鼠肾脏组织MDA和ROS含量较DM组均有不同程度的降低,而SOD活性均有显著升高,其中Tau+Pue组这种变化更加明显;光学显微镜下显示:DM组大鼠肾小球毛细血管管腔闭塞,系膜细胞增多,基膜增厚和系膜基质大量堆积,肾小球体积增大。肾小管明显扩张,近曲小管上皮细胞肿胀、变性;牛磺酸联合葛根素处理组大鼠肾脏上述损伤性变化明显减轻。结论牛磺酸联合葛根素对2型糖尿病大鼠肾脏有保护作用,其机制可能与降低氧化应激水平有关。
Objective To observe the protective effects of taurine and puerarin on the kidney of type 2 diabetic rats. Methods Forty Wistar rats were randomly divided into four groups: diabetic group (DM group), taurine group (Tau group), Puerarin alone group (Pue group) and taurine plus Puerarin group group). After 8 weeks, the plasma glucose, creatinine (BUN), superoxide dismutase (SOD), malondialdehyde (MDA), reactive oxygen species (ROS) Insulin and renal tissue MDA, ROS and SOD were measured. The renal index was calculated and the pathological changes of kidney were observed under light microscope. Results Compared with DM group, plasma SOD activity and insulin level were increased in Tau group, Pue group and Tau + Pue group, while Glucose, Cre, BUN, MDA, ROS and renal index were decreased to some extent, The content of MDA and ROS in the kidney of the drug-treated rats were all decreased to some extent compared with the DM rats, while the activity of SOD was significantly increased, especially in the Tau + Pue group. Under optical microscope, Small capillaries lumen occlusion, mesangial cells increased thickening of the basement membrane and a large number of accumulation of mesangial matrix, glomerular volume increased. Tubular expansion, proximal tubule epithelial cell swelling, degeneration; taurine combined with puerarin treatment group rats kidney damage significantly reduced. Conclusion Taurine combined with puerarin has a protective effect on the kidney of type 2 diabetic rats. The mechanism may be related to the reduction of oxidative stress.
