目的:通过对ABO疑难血型的血清学与基因检测分析,探讨ABO亚型的分布状况和ABO血型基因分型的意义。方法:采用试管法正反定型、吸收放散试验等一系列血型血清学方法检测分析97例正反定型不符血液标本的血清学表型,对其中61例标本,辅以聚合酶链式反应-特异性序列引物(PCR-SSP)基因分型技术检测其基因型。结果:97例ABO疑难样本中检出A亚型13例,B亚型8例,AB亚型50例,B(A)18例、CisAB 5例,Bh m 1例,O型抗-B抗体缺失1例,1例有直抗阳性和自身抗体干扰,排除后被定为A型。PCR-SSP技术检出常见ABO基因4个(A、A201、B、O),以及ABO基因型6个(A/O、A201/O、B/O、A/B、A201/B、O/O)。结论:ABO疑难血型的鉴定应采用多种血清学方法进行检测,PCR-SSP技术能识别ABO基因型,作为一种辅助方法可应用于检测ABO疑难血型。
OBJECTIVE: To investigate the distribution of ABO subtypes and the significance of ABO genotypes by analyzing serological and genetic analysis of ABO difficult blood types. Methods: Serological phenotypes of 97 cases of non-conforming and non-conforming blood samples were detected and analyzed by a series of blood group serological methods, including positive and negative test-tube assay, absorption and desorption test, and 61 of them were supplemented with polymerase chain reaction-specific Genotypes were detected by PCR-SSP genotyping. Results: Among the 97 samples of ABO, 13 cases were identified as A subtype, 8 subtype B, 50 subtypes of AB, 18 of B (A), 5 of CisAB, 1 of Bh m, One case was deleted, one case had direct anti-positive and autoantibody interference, and was excluded as type A after exclusion. Four ABO genes (A, A201, B, O) and six ABO genotypes were detected by PCR-SSP technique. Six ABO genotypes were identified as A / O, A201 / O). Conclusion: The identification of ABO difficult blood type should be detected by many serological methods. The PCR-SSP technique can identify ABO genotype and can be used as an auxiliary method to detect ABO difficult blood type.
