目的观察体外培养的成人毛囊隆起细胞的生长特性。方法采用一步消化法和显微分割技术获取毛囊隆起细胞,并进行培养、传代,连续观察细胞的生长状况、形态特征以及表达特性。结果原代培养的第2d便可见细胞生长,并表现出很强的增殖潜能,6d时达到最高,并维持数日;光镜下细胞小而圆,大小一致,可见有丝分裂相;免疫细胞化学染色可见K19阳性细胞,表达随培养时间延长呈下降趋势。结论体外培养的毛囊隆起细胞具有原始细胞特性,提示毛囊中潜在的干细胞定位于毛囊隆起。
Objective To observe the growth characteristics of adult hair follicle bump cells cultured in vitro. Methods The hair follicle cells were obtained by one-step digestion and microdissection. The cells were cultured, passaged, and continuously observed for cell growth status, morphological characteristics and expression characteristics. Results The cells were seeded on the 2nd day of primary culture and showed a strong proliferative potential, which reached the highest level on the 6th day and maintained for several days. The cells were small and round under the light microscope with the same size and mitosis. Immunocytochemical staining Visible K19 positive cells, the expression showed a downward trend with the incubation time. Conclusion The hair follicle cells cultured in vitro have the characteristics of primitive cells, suggesting that the potential stem cells in the hair follicles are located in the hair follicles.
