目的通过观察昆明小鼠心脏发育过程中Klotho基因的定位及其表达。方法新生1、180、540 d昆明小鼠各5只,断颈处死,分别取出肾脏和心脏,并将心脏的右心房部分剪下;1、2、4、7、15、30、60、120、180、270、360 d和540 d昆明小鼠各10只,断颈处死后取出心脏组织;以上均提取总RNA,应用荧光定量RT-PCR技术检测Klotho mRNA的表达。结果切除右心房的心脏组织,未能检测到Klotho mRNA的表达,肾脏和右心房存在表达;昆明小鼠出生后即可在心脏检测到Klotho mRNA的表达,1、2、4、7、15、30、60、120 d小鼠相对表达量基本呈上升趋势;270、360、540 d小鼠相对表达量基本呈下降趋势。结论 Klotho基因在昆明小鼠的心脏组织中可能仅在右心房表达;120 d之内,Klotho基因在心脏组织的表达量有随增龄而增加的趋势;270 d之后,Klotho基因在心脏组织的表达量有随增龄而减弱的趋势。
Objective To observe the localization and expression of Klotho gene in the process of cardiac development in Kunming mice. METHODS: Five newly born Kunming mice (1,180 and 540 days) were sacrificed and their necks and hearts were sacrificed. The right atrium of the heart was partially excised. 1,2,4,7,15,30,60,120 , 180, 270, 360 d and 540 d Kunming mice. The heart tissues were removed after the neck was ablated. The total RNA was extracted and the expression of Klotho mRNA was detected by real-time fluorescence quantitative RT-PCR. Results The cardiac tissue of the right atrium was resected, and the expression of Klotho mRNA was not detected in the kidneys and the right atrium. Klotho mRNA expression was detected in the heart of Kunming mice at 1, 2, 4, 7, 15, The relative expression level of mice in 30,60 and 120 days increased gradually. The relative expression level in 270,360 and 540 days mice showed a decreasing trend. Conclusions Klotho gene may be expressed only in the right atrium of Kunming mouse heart tissue. Within 120 days, Klotho gene expression in heart tissue tends to increase with age. After 270 days, Klotho gene expression in heart tissue The expression of the trend with age and weakened.
