目的探讨石棉暴露对接触人群血浆micro RNA(mi RNA)表达的影响。方法选取暴露组(暴露年限>10年)和对照组(未暴露)各3人,用mi RNA芯片比较两组对象血浆mi RNA表达差异,使用Target Scan、mi Randa和Pic Tar 3种在线软件对差异表达mi RNA进行靶基因预测,最后对靶基因进行基因本体论分析和全基因组及代谢途径数据库通路分析。结果 mi RNA芯片筛测结果显示,石棉暴露人群血浆中40个mi RNA的表达与对照组存在差异(P<0.05),其中有9个mi RNA信号值>500。对这些信号值>500的mi RNA的靶基因进行预测和通路分析,有5个mi RNA检索到相应的靶基因,这些基因主要参与肿瘤相关的信号通路和代谢通路,包括FGFR3等潜在功能靶点。结论石棉暴露可致接触人群血浆中mi RNA发生差异表达,其靶基因主要参与肿瘤相关的信号通路,但其在石棉相关疾病中的作用机制有待进一步研究。
Objective To investigate the effect of asbestos exposure on plasma micro RNA (miRNA) expression in exposed population. Methods Three miRNAs were selected from the exposed group (exposed for more than 10 years) and the control group (unexposed), respectively. The difference of plasma mi RNA expression between the two groups was compared using mi RNA microarray. Three online softwares, Target Scan, mi Randa and Pic Tar Differentially expressed mi RNAs were used to predict target genes. Finally, gene ontology analysis and genome-wide and metabolic pathway database analysis of target genes were performed. Results The results of mi RNA microarray showed that the miRNA expression in 40 plasma exposed to asbestos was significantly lower than that in the control group (P <0.05). Among them, 9 miRNA signals were> 500. To predict and analyze the target genes of miRNAs with signal> 500, five miRNAs were found to target the corresponding target genes. These genes are mainly involved in tumor-related signaling pathways and metabolic pathways, including potential functional targets such as FGFR3 . Conclusion The miRNAs in the plasma of exposed human asbestos exposed to asbestos are differentially expressed. The target genes are mainly involved in tumor-related signaling pathways. However, their role in asbestos-related diseases remains to be further studied.
