目的为了阐明凝血酶对肾小球系膜细胞细胞间粘附分子１（ＩＣＡＭ－１）表达的影响。方法利用体外培养的人肾小球系膜细胞，分别采用噻唑蓝掺入法、流式细胞术、间接免疫荧光、Ｎｏｒｔｈｅｒｎ杂交等方法分别观察了凝血酶对系膜细胞增殖和表达ＩＣＡＭ－１蛋白和ｍＲＮＡ的影响，并同时进行了人工重组水蛭素的阻断实验。结果凝血酶可以显著促进系膜细胞增殖以及系膜细胞ＩＣＡＭ－１蛋白和ｍＲＮＡ的表达，水蛭素可以特异地阻断凝血酶的这些作用。结论凝血酶不仅是肾小球内凝血过程的关键酶，而且具有促进系膜增殖和促进系膜细胞表达ＩＣＡＭ－１的作用。
Objective To elucidate the effect of thrombin on the expression of intercellular adhesion molecule-1 (ICAM-1) in glomerular mesangial cells. Methods Human glomerular mesangial cells cultured in vitro were respectively treated with thiazolyl tetrazolium (MTT), flow cytometry, indirect immunofluorescence and Northern blotting to observe the effects of thrombin on the proliferation of mesangial cells and the expression of ICAM-1 protein And mRNA effects, and at the same time artificial recombinant hirudin blocking experiment. Results Thrombin can significantly promote the proliferation of mesangial cells and the expression of ICAM-1 protein and mRNA in mesangial cells. Hirudin can specifically block these effects of thrombin. Conclusion Thrombin is not only the key enzyme in glomerular coagulation process, but also has the effect of promoting mesangial proliferation and promoting the expression of ICAM-1 in mesangial cells.
