目的 构建呈现肿瘤相关抗原P815A的噬菌体颗粒 ,在体诱导针对肥大细胞瘤P815的特异性细胞毒性T细胞 (CTL)反应。方法 构建呈现CTL表位P1A35~ 4 3 的噬菌粒载体 ,大量制备呈现此表位的重组噬菌体颗粒 ,小剂量、无佐剂皮下免疫DBA/2小鼠 (H 2 d) ,采用51Cr释放法、ELISPOT观察针对肥大细胞瘤P815的CTL反应。结果 重组噬菌体呈现颗粒疫苗在体诱导了针对弱抗原P815A的CTL反应。结论 丝状噬菌体呈现疫苗增强肿瘤相关抗原的免疫原性 ,是一种颇有潜力的抗肿瘤疫苗形式
Objective To construct a phage particle that displays tumor-associated antigen P815A and induce a specific cytotoxic T cell (CTL) response against mast cell tumor P815 in vivo. Methods A phagemid vector containing CTL epitopes P1A35-43 was constructed, and a large number of recombinant phage particles presenting this epitope were prepared. Small doses of subcutaneously immunized DBA/2 mice (H 2 d) were prepared using the 51Cr release method. , ELISPOT observation of CTL response to mast cell tumor P815. Results Recombinant phage display particle vaccine induced in vivo CTL response against weak antigen P815A. Conclusion Filamentous bacteriophage presenting vaccine enhances the immunogenicity of tumor-associated antigens. It is a potential anti-tumor vaccine form.
