目的:考察温度及pH对甘草酸溶液中内毒素团聚状态的影响。方法:采用30 kDa Millipore膜过滤不同温度及pH条件下甘草酸溶液,通过动态浊度法测定超滤液中内毒素活性浓度,HPLC检测超滤前后有效成分的变化,利用内毒素纳米粒径仪测定不同温度及pH条件下内毒素团聚状态的粒径分布。结果:温度60℃,pH 4时,内毒素团聚状态的粒径分布和去除率均最大,甘草酸溶液经30 kDa Millipore膜超滤前后有效成分含量>98%。结论:随温度的升高,甘草酸溶液中内毒素团聚状态的粒径分布变大,而随pH的增大则变小。
Objective: To investigate the effects of temperature and pH on the agglomeration of endotoxin in glycyrrhizic acid solution. Methods: The 30 kDa Millipore membrane was used to filtrate the glycyrrhizic acid solution under different temperature and pH conditions. The concentration of endotoxin in the ultrafiltrate was determined by dynamic turbidity method. The changes of active components before and after ultrafiltration were detected by HPLC. The particle size distribution of the agglomeration of endotoxin under different temperature and pH conditions was determined. Results: At 60 ℃ and pH 4, the particle size distribution and removal rate of the endotoxin in the agglomeration state were the highest. The contents of active ingredients in the solution of glycyrrhizic acid before and after ultrafiltration with 30 kDa Millipore membrane were> 98%. CONCLUSION: With the increase of temperature, the particle size distribution of LPS in glycyrrhizic acid solution becomes larger, but decreases with the increase of pH.
