目的旨在从基因转录水平确定培养的人晶状体上皮细胞是否具有表达ⅠⅢ型胶原的生物学特性。方法采用人眼晶状体囊膜进行晶状体上皮细胞体外的原代培养;细胞融合后,抽提RNA,应用Northernblot分析技术进行探测人晶状体上皮细胞的基因转录产物。结果人晶状体上皮细胞的基因转录产物中存在有Ⅰ、Ⅲ型前胶原mRNA的表达,表达的二型前胶原mRNA均存在有长度为5.8、5.4、4.8kb的片段,Ⅰ型前胶原mRNA表达的量是血型前胶原mRNA的3倍。结论培养的人晶状体上皮细胞能够合成Ⅰ、Ⅲ型胶原,所合成的这二型胶原中,以Ⅰ型为主,它们可能对后囊膜混浊的形成起到重要的作用。
The aim is to determine from the gene transcriptional level whether cultured human lens epithelial cells have the biological characteristics of expressing type III collagen. Methods Human lens epithelial cells were primary cultured in vitro by human lens capsule. After the cells were fused, RNA was extracted and Northern blot analysis was used to detect the gene transcription products of human lens epithelial cells. Results The expression of type Ⅰ procollagen and type Ⅲ procollagen mRNA were found in human lens epithelial cells. The expression of type Ⅱ procollagen mRNA was found in 5.8, 5.4 and 4.8 kb fragments, The amount of procollagen mRNA expression is three times higher than that of blood pre-procollagen mRNA. Conclusion The cultured human lens epithelial cells can synthesize type I and type III collagen. The type I collagen of these two types of collagen is mainly type I, which may play an important role in the formation of posterior capsule opacity.
