目的:研究化浊解毒活血通络方对脑缺血再灌注损伤大鼠神经功能评分及Caspase-3表达的影响,探讨其神经保护作用机制。方法:采用改良线栓法建立大鼠脑缺血再灌注损伤模型。运用Longa法进行神经病学评分,用光镜观察脑组织形态学改变,用PCR法检测Caspase-3的表达。结果:模型组及各用药组均出现不同程度的神经功能缺损,模型组Caspase-3表达明显多于假手术组(P<0.05);与模型组相比,中药各组及阿司匹林组组织变性坏死程度轻、Caspase-3阳性细胞减少(P<0.05),中药高剂量组及阿司匹林组神经功能缺损评分显著降低(P<0.05);中药中、高剂量组和阿司匹林组效果优于中药低剂量组(P<0.05)。结论:化浊解毒活血通络方可以通过抑制Caspase-3的阳性表达来抑制细胞凋亡,从而发挥其神经保护作用。
Objective: To investigate the effects of Huazhuo Jiedu Huoxue Tongluo Recipe on neural function score and expression of Caspase-3 in rats with cerebral ischemia-reperfusion injury and to explore its neuroprotective mechanism. Methods: A rat model of cerebral ischemia-reperfusion injury was established by modified suture method. The neurological score was assessed by Longa’s method. Morphological changes of brain tissue were observed by light microscope. Caspase-3 expression was detected by PCR. Results: Compared with the model group, the expression of Caspase-3 in the model group and the treatment groups were significantly different (P <0.05). Compared with the model group, the changes in the tissue of the Chinese medicine group and the aspirin group were degenerative and necrotic The level of Caspase-3 positive cells decreased (P <0.05), and the score of neurological deficit in Chinese medicine high-dose group and aspirin group decreased significantly (P <0.05); Chinese medicine high-dose group and aspirin group were better than low dose Chinese medicine group (P <0.05). Conclusion: Huazhuo Jiedu Huoxue Tongluo Recipe can inhibit apoptosis by inhibiting the positive expression of Caspase-3, and thus exert its neuroprotective effect.
