目的观察石榴叶总酚对小鼠脑缺血再灌注损伤的保护作用,并研究其可能的作用机理。方法石榴叶总酚按照60,120和240 mg·kg-1给小鼠灌胃3 d后,第4天给药1 h后,采用双侧颈总动脉反复阻断再通造成小鼠脑缺血再灌注损伤模型,采用水迷宫实验检测术后不同时间小鼠学习记忆能力的变化。观察术后24 h内脑组织中MDA含量和SOD活性的变化以及相关炎性因子的表达。结果石榴叶酚可以显著改善脑缺血再灌小鼠学习记忆能力,降低脑组织内MDA含量,提高SOD活性,明显降低IL-1β,TNFα,COX2mRNA的表达,同时下调TLR2和NOD2mRNA的表达。结论石榴叶酚灌胃给药对双侧颈动脉缺血再灌注引起的小鼠学习记忆能力降低有明显的改善作用,该作用与其抗氧化和抑制局部脑组织内前炎性相关因子表达和固有免疫系统TLR2和NOD2的表达有关。
Objective To observe the protective effect of total phenolics of pomegranate leaf on cerebral ischemia-reperfusion injury in mice and to study its possible mechanism. Methods The total phenolics of pomegranate leaf were administered to mice at 60, 120 and 240 mg · kg-1 for 3 days. The rats were administered with Bilateral common carotid artery for 1 h. Perfusion injury model, water maze test was used to detect the changes of learning and memory ability of mice at different time points. The changes of MDA content and SOD activity and the expression of related inflammatory factors in brain tissue were observed within 24 hours after operation. Results pomegranate leaf phenolics can significantly improve the learning and memory ability, reduce the content of MDA in brain tissue and increase the activity of SOD, decrease the expressions of IL-1β, TNFα and COX2 mRNA, and down-regulate the expressions of TLR2 and NOD2 mRNA in mice with cerebral ischemia-reperfusion. Conclusion Galectin-gavage administration significantly reduces the learning and memory abilities of mice induced by bilateral carotid artery ischemia-reperfusion, which is related to its anti-oxidation and anti-inflammatory effects and the inhibition of the expression of intrinsic proinflammatory cytokine The immune system TLR2 and NOD2 expression.
