采用摇瓶培养方法,通过单因素试验研究发酵培养液初始p H值、装液量、接种量和摇床转速对桑黄(Phellinus linteus)发酵胞内黄酮产量的影响,结果表明,最适初始p H为6.5,装液量为100 m L/300m L,接种量为10%,摇床转速为150 r·min~(-1)。在单因素试验的基础上采用L9(34)正交试验优化桑黄黄酮发酵培养条件,最佳工艺为:发酵液初始p H6.5,300 m L三角瓶装液量为100 m L,种子液接种量为10%,摇床转速150 r·min~(-1)。
The effects of initial pH, liquid volume, inoculum size and shaking speed on the intracellular flavonoid production of Phellinus linteus were studied by single-factor experiments in shake flask culture. The results showed that the optimal initial p H was 6.5, the volume of liquid was 100 m L / 300 m L, inoculation amount was 10%, and the shaking speed was 150 r · min -1. On the basis of single factor experiment, orthogonal experiment L9 (34) was used to optimize the fermentation conditions of Phellinus yellow flavone. The optimum conditions were as follows: the initial p H6.5 of the fermentation broth was 300 ml, the volume of the flask was 100 m L, 10%, shaking speed 150 r · min ~ (-1).
