为了解硝苯吡啶对血管平滑肌细胞连接通讯的影响,采用荧光黄划痕示踪法,测定硝苯吡啶作用的培养的大鼠平滑肌细胞连接通讯功能,并通过显微荧光光度计进行定量分析。此外,采用免疫组织化学S-P法了解硝苯吡啶对该类细胞连接蛋白43表达的影响,通过计算机图像分析进行定量研究。结果显示,硝苯吡啶促进细胞连接蛋白43的表达和细胞连接通讯。提示硝苯吡啶通过促进细胞连接蛋白43的表达和细胞连接通讯来抑制平滑肌细胞增殖,是抗动脉粥样硬化作用的重要机制之一。
In order to investigate the effect of nifedipine on the vascular smooth muscle cell junctions and communication, the fluorescent-yellow scratch-tracing method was used to determine the nifedipine-mediated cultured rat smooth muscle cell connectivity and quantitative analysis by micro-fluorophotometer. In addition, immunohistochemical S-P method to understand the effect of nifedipine on the expression of this type of cell connexin 43, quantitative analysis by computer image analysis. The results showed that nifedipine promoted the expression of connexin43 and cell ligation. It is indicated that nifedipine inhibits the proliferation of smooth muscle cells by promoting the expression of connexin 43 and cell-cell communication, which is one of the important mechanisms of anti-atherosclerosis.
