目的:通过多因素正交试验法优选双频复合超声提取丹参的工艺,为规范丹参的提取工艺提供技术参数。方法:采用正交试验法,以丹参酮ⅡA为指标成分考察了乙醇浓度、物料粒径、固液比、提取温度、提取时间5个因素,对丹参的超声提取工艺进行优选。结果:各因素对丹参中丹参酮ⅡA提取率的影响大小为:乙醇浓度>物料粒径>固液比>提取时间>提取温度。最佳提取工艺为乙醇浓度90%,物料粒径80~100目,固液比1∶20,提取时间35min,提取温度55℃。在此条件下,丹参酮ⅡA的提取率是89.47%。结论:本研究将为丹参工业化生产提供技术参数。
OBJECTIVE: To optimize the extraction process of Salvia miltiorrhiza by dual-frequency composite ultrasound and to provide technical parameters for the standard extraction of Salvia miltiorrhiza. Methods: Orthogonal test was used to study the ultrasonic extraction process of Salvia miltiorrhiza Bunge by using tanshinone ⅡA as the index component to study the factors of ethanol concentration, material size, solid-liquid ratio, extraction temperature and extraction time. Results: The effects of various factors on the extraction rate of tanshinone ⅡA in Salvia miltiorrhiza were: ethanol concentration> material particle size> solid-liquid ratio> extraction time> extraction temperature. The best extraction process was ethanol concentration 90%, material size 80 ~ 100 mesh, solid-liquid ratio 1:20, extraction time 35min, extraction temperature 55 ℃. Under these conditions, the extraction rate of tanshinone ⅡA was 89.47%. Conclusion: This study will provide technical parameters for Salvia miltiorrhiza industrial production.
