目的:观察槟榔碱对上皮细胞Hacat凋亡的影响。方法:用0,25,50,75,100,125μg/mL槟榔碱刺激上皮细胞Hacat24h,抽提DNA进行琼脂糖凝胶电泳和流式细胞术检测。结果:100和125μg/mL槟榔碱刺激后上皮细胞Hacat DNA梯状条带明显;50,75,100,125μg/mL槟榔碱诱导上皮细胞Hacat凋亡并呈剂量依赖性,随浓度升高进入晚期凋亡细胞的比例上升。结论:槟榔碱能通过诱导上皮细胞凋亡参与口腔黏膜下纤维性变的发生。
Objective: To observe the effects of arecoline on apoptosis of Hacat cells in epithelial cells. Methods: Hacat cells were stimulated with arecoline at concentrations of 0, 25, 50, 75, 100 and 125μg / mL for 24 hours. DNA was extracted and analyzed by agarose gel electrophoresis and flow cytometry. Results: The Hacat DNA ladder was observed in the epithelial cells stimulated with arecoline 100 and 125 μg / mL. 50, 75, 100, 125 μg / mL arecoline significantly induced Hacat apoptosis in a dose - dependent manner. The proportion rose. CONCLUSION: Arecoline can participate in the occurrence of oral submucous fibrosis by inducing epithelial cell apoptosis.
