目的观察加味左归饮对绝经后骨质疏松大鼠下丘脑雌激素受体α(ERα)及其基因表达的影响。方法以切除卵巢的雌性大鼠作为骨质疏松的动物模型,将大鼠分为假手术组、模型组、阳性对照组和加味左归饮低、中、高剂量组,用免疫组织化学方法和原位杂交方法检测下丘脑ERα及其mRNA表达。结果模型组ERα受体及其mRNA阳性细胞与假手术组比较显著减少;而加味左归饮能够提高去卵巢后下丘脑ERα及其mRNA阳性细胞数。结论加味左归饮可通过增加去卵巢大鼠下丘脑ERα表达以增强雌激素的生物效应。
Objective To observe the effect of Modified Zuo Gui Yin on the expression of estrogen receptor α (ERα) and its mRNA in hypothalamus of postmenopausal osteoporosis rats. Methods Ovariectomized female rats were used as an animal model of osteoporosis. The rats were divided into sham operation group, model group, positive control group and low-, middle-, and high-dose group of modified Zuo Gui Yin. In situ hybridization was used to detect the expression of ERa and its mRNA in the hypothalamus. Results Compared with the sham-operated group, the ERα receptor and its mRNA positive cells in the model group were significantly decreased. However, modified Zuoguiyin can increase the number of ERα and its mRNA positive cells in the hypothalamus after ovariectomy. [Conclusion] Modified Zuo Guiyin can enhance the biological effects of estrogen by increasing the expression of ERα in the hypothalamus of ovariectomized rats.
