目的 探讨wortmannin抗眼内新生血管形成的可能。方法 采用免疫荧光、流式细胞分析和Boyden小室迁移实验观察高糖视网膜Müller细胞条件培养基(HGMCM)诱导的内皮细胞增殖和迁移的影响。结果 50nmol/Lwortmannin可明显抑制HGMCM诱导的内皮细胞的增殖和迁移,使内皮细胞S期细胞百分率明显下降[ (37 .82±0 .57)%至(21. 91±0 .23)%,P<0 .01],G0 /G1 期细胞百分率显著升高[ (54 .57±1 .19)%至(65. 59±0. 41)%,P<0 .01],G2 /M期细胞增多(P<0 .05)。结论 wortmannin对HGMCM诱导的内皮细胞的增殖和迁移均有抑制作用,提示wortmannin具有抗视网膜新生血管形成的作用。
Objective To investigate the possible anti-ocular neovascularization of wortmannin. Methods Immunofluorescence, flow cytometry and Boyden chamber migration assay were used to observe the effects of HGMCM on the proliferation and migration of endothelial cells. Results 50 nmol / L wortmannin significantly inhibited the proliferation and migration of endothelial cells induced by HGMCM, and significantly decreased the percentage of S phase cells in endothelial cells [(37.82 ± 0.57)% vs (21.91 ± 0.23)%, P <0. 01]. The percentage of cells in G0 / G1 phase was significantly higher than that in G2 / M phase [(54.57 ± 1.19)% vs (65.59 ± 0.41)%, P <0.01] Increased (P <0 .05). Conclusion wortmannin inhibits the proliferation and migration of endothelial cells induced by HGMCM, suggesting that wortmannin may have anti-retinal neovascularization effects.
