通过HPLC法比较湘西宏成制药有限责任公司不同中药种子繁殖基地白花泡桐叶中熊果酸和木犀草素的含量。所采用的色谱柱为Shim-Pack C18(4.6 mm×150 mm,5μm),测定熊果酸的流动相为甲醇-水-乙酸(85∶15∶0.5),流速为1.0 mL/min,检测波长210 nm;测定木犀草素的流动相为甲醇-水-乙酸(55∶44∶1),流速为1.0 mL/min,检测波长350 nm。试验表明,熊果酸在1~15μg范围内线性关系良好,回归方程为Y=0.020 6 X-0.466 6,R=0.999 0,平均回收率为96.1%,RSD为1.47%;木犀草素在0.1~1μg范围内线性关系良好,回归方程Y=0.020 1 X+0.002 0,R=0.999 0,平均回收率为96.5%,RSD为1.65%。检测结果表明,宏成公司4号基地白花泡桐叶中熊果酸和木犀草素的含量显著高于其他3个基地白花泡桐。试验为白花泡桐药材质量标准的建立提供理论依据。
The contents of ursolic acid and luteolin in leaves of Paulownia fortunei from different traditional Chinese medicine seed multiplication bases were compared by HPLC. The chromatographic column was Shim-Pack C18 (4.6 mm × 150 mm, 5 μm). The mobile phase of ursolic acid was methanol-water-acetic acid (85:15:0.5) and the flow rate was 1.0 mL / min. 210 nm. The mobile phase of luteolin was methanol-water-acetic acid (55:44:1), the flow rate was 1.0 mL / min and the detection wavelength was 350 nm. The results showed that ursolic acid had a good linearity in the range of 1 ~ 15μg, the regression equation was Y = 0.020 6 X-0.466 6, R = 0.999 0, the average recovery was 96.1% and the RSD was 1.47% The regression equation was Y = 0.020 1 X + 0.002 0, R = 0.999 0 with an average recovery of 96.5% and a RSD of 1.65%. The results showed that the content of ursolic acid and luteolin in the leaves of Hongchengbaoyong, a base of Hongcheng Company, was significantly higher than that of the other three bases.
