目的探讨哮喘血清、地塞米松、白细胞介素-4(IL-4)、干扰素γ(IFN-γ)、转化生长因子-β(TGF-β)等不同刺激因子对人气道平滑肌细胞白细胞介素-22R1(IL-22R1)mRNA及蛋白表达的影响。方法用免疫荧光PCR检测不同因素刺激下人气道平滑肌细胞IL-22R1mRNA的表达,用Westernblot检测人不同因素刺激下的人气道平滑肌细胞IL-22R1蛋白的表达,比较不同组别之间的差异。结果哮喘血清刺激下的人气道平滑肌细胞IL-22R1mRNA升至对照组345倍,而蛋白表达明显增强(P<0.01);加用地塞米松后IL-22R1mRNA降至对照组10倍,蛋白表达较仅用哮喘血清刺激明显减弱(P<0.01)。IL-4、IFN-γ、TGF-β刺激后人气道平滑肌细胞IL-22R1mRNA及蛋白表达均显著性增加。结论 IL-22R1表达的改变可能参与支气管哮喘的病程,影响人气道平滑肌细胞IL-22R1受体的表达可能是地塞米松发挥其在支气管哮喘病程中作用的机制之一。IFN-γ、IL-4、TGF-β对气道平滑肌细胞IL-22R1的作用可能是其对支气管哮喘疾病产生影响的机制之一。
Objective To investigate the effects of asthmatic serum, dexamethasone, interleukin-4 (IL-4), interferon-γ (IFN-γ) and transforming growth factor-β (TGF-β) on airway smooth muscle cell leukocyte Effects of IL-22R1 mRNA and protein expression on the expression of IL-22R1. Methods The expression of IL-22R1 mRNA in human airway smooth muscle cells stimulated by different factors was detected by immunofluorescence PCR. The expression of IL-22R1 protein in human airway smooth muscle cells stimulated by different human factors was detected by Western blot, and the differences among different groups were compared. Results IL-22R1mRNA of human airway smooth muscle cells stimulated by asthmatic serum increased to 345-fold and the protein expression was significantly increased (P <0.01), while the expression of IL-22R1 mRNA decreased 10-fold in control group after dexamethasone administration Stimulation with asthma serum was significantly attenuated (P <0.01). IL-4, IFN-γ, TGF-β stimulation of human airway smooth muscle cells IL-22R1 mRNA and protein expression were significantly increased. Conclusion The change of IL-22R1 expression may be involved in the course of bronchial asthma. It may be one of the mechanisms of dexamethasone exerting its role in the process of bronchial asthma by affecting the expression of IL-22R1 receptor in human airway smooth muscle cells. The effects of IFN-γ, IL-4 and TGF-β on IL-22R1 in airway smooth muscle cells may be one of the mechanisms that affect the disease of bronchial asthma.
